I think this is Martin Brisby from The Secret of NIMH movies.

Yes, this is Martin, the spoiled brat from The Secret of NIMH, then the usurper from Timmy To The Rescue. Voiced by Wil Wheaton, pre-ST:TNG. Good deducing, Glow.

Saw a picture of him all evil once floating around Devianart--got curious and looked him up. Knowledge simply came in handy later.
Here's one that simply had to be done.
1. We're a pair of homely females.
2. While looks aren't everything, we aren't very nice either.
3. One of us actually does get redeemed in the two direct to video sequels--the other one of us is still just as nasty.

These sound like Lady Tremaine's daughters, Hortense and Prudence, from Disney's Cinderella and its sequels.

Actually, their names are Anastasia and Drizella.

Correct, their names are actually Anastasia and Drizella--Drizella being the brunette in green and Anastasia being the redhead in pink--who gets redemption in the 2nd and 3rd direct to video sequels. Drizella on the other hand remains cruel and nonredeemable.

I made the clues simple enough, or so I thought. At any rate, here are the last three clues:

This is Gaetan Moliere, known simply as "Mole" for his fixation on soil and earth samples. He's part of Preston Whitmore's hand-picked team to find the lost city of Atlantis, especially because nobody digs like Moliere, not even Gaston.

Okay, speaking of Mole...

1) This character occurs only once to date in a partly animated feature film. It's a Space Jam kind of thing going on, because Hollywood is hopeless.

2) The Smurfette Principle is in effect: this character is the only female on a team of males. Two of her teammates debate about which one has her fancy.

3) While this character is not a mole herself, she is acquainted with one.

This is Juarez from G-Force--the Penelope Cruz voiced guinea pig.

That's her, Juarez the guinea pig from Disney Pictures' G-Force. She keeps Darwin and Blaster going in circles for her affections. One other member of the team is Speckles the mole, who's the tech wizard. Good memory, GW.

Had to look up her name--but sadly I remember those advertisements for that movie.

2. I'm a bit of a fanboy--nothing this guy's ever done is wrong in my eyes--even when it clearly is.
3. However, in the recent live action version--it's more than just a man-crush and I actually do switch sides near the end.

This is Lefou, from Beauty and the Beast.

Correct. From what I read, in the recent live action movie, he has an unrequited love for Gaston and he actually joins Belle and the Castle staff on the fight as Gaston is too far gone by now.
